Bead Enrichment Protocol

In this protocol, 3 um diameter, low density, non-magnetic polystyrene beads are pre-loaded with a biotinylated capture oligo. A suspension of the amplified and unamplified ePCR beads is added, with the amplified beads hybridizing to the capture oligos on the low-density beads. The mixture is then centrifuged to separate unamplified ePCR beads from the amplified-bead / capture-bead complexes. The low-density complexes remain in the supernatant, while the unamplified beads form a pellet. The supernatant is recovered, the complexes are melted with NaOH, and the enriched, amplified beads are recovered by magnetic separation.
