Emulsion PCR Protocol
Monosized, paramagnetic, streptavidin-coated MyOne beads are pre-loaded with a dual biotin
forward primer. An aqueous phase is prepared with the above beads, low-cost PCR
polymerase, PCR buffers and dNTPs, and template DNA. This is mixed with an oil phase, and
vortexed to create the emulsion. Each droplet will ideally have one bead, and one molecule
of template DNA. PCR is then performed to amplify the template DNA within each emulsion
droplet. A simple device to implement the emulsion PCR protocol will be available for sale in
2009.
